Hmmm...my best advice on sequential art would be to make sure the pictures and the dialogue really match up, I know it does seem like a pretty obvious thing but I've seen some very good comics be rendered unreadable by crazy text layout (For example a comic I read that was to be read left-to-right had the first speaker's text at the bottom and to the right, which made it hard to "hear" what the characters were saying:(not actual quote) "You never have anything under control!" "don't worry, I have this under control!" Just make sure the first speaker's text is always higher.
Now that we're over that part, panel layout. Will you be using traditional methods or a computer program? For pen and ink, I suggest a ruler (I used to shun the ruler and still do largely but if I'm going to do a panel border I have to give) A good thing to keep in mind is what you are going to do in the panels. It doesn't make much sense to put a small drawing in a big box with minimal text, any more than it does covering up artwork you slaved over with too much text. This is wher you'll need to keep track of the pace of the comic. Say, a character needs to make some dramatic speech or something. Usually an artist will do what is call a "talking head" panel, where it is usually just an upper torso, a disembodied head or other such room-saving technique to represent the talking character. You don't want to have text dominate a panel, if anything I'm sure people prefer a silent art panel to a panel of pure conversation, unless the dialogue itself is the best part hehe.
Timing is key in a comic, of you sell the joke too soon it may fall flat and if you draw it out too long people may lose interest before they get to the punchline. Don't go out of your way to explain a joke either, if the audience doesn't get it just consider yourself smarter than them heheh (naw that's kinda mean.) But you may have to assume a more general volcabulary. For example, I had a joke about a buzzard in an airport. The buzzard was confused because they wouldn't allow him on with a carry-on bag. He misunderstood it for "carrion" (dead animal carcasses, yum!) but in one of the initial writes, I had the word "incredulous" in there, assuming my friends knew what it meant.A quick ask around the table of about 13 at Denny's blew a hole in that theory, no one had even heard of the word "incredulous" so the word became "befuddled", an easier to understand yet somehow more intelligent sound alternative to "puzzled" And that's a good thing too, be o[en to alternatives to jazz speech up a little without using big obnoxious words.
As for panels, the only true purpose is to seperate the art, so there should be no worry as to where those go. I'll talk more about this in a bit... any questions and I'll be glad to help, I'm sure there are others that have very good ideas too that they can share ^_^

If I may add... Well... This may not be correct.... But I HAVE seen it in several manga:
Breaking the panel borders. This is fine, it helps the page feel looser. If all of your characters are crammed into bxy little panels, it might get old. Many people do very wel with the little panel style, but even a braid flying out of the panel makes the page more alive.
Layout. This is one of the hardest parts. You need to have a varieaty of panel types, such as rhombi, triangles, irregular shapes. I beleive that the majority of panels should be square or rectangular, but I think that different kinds of shapes will help the action flow. Also, if you are doing a full shot of a characer, you could leave the panel out entirely!
Characters: It doesn't seem like you need my help on this, you probably don't need my help on anything, but characters is one of my favorite parts of comic writing. Firat, I would suggest drawing a few people, trying to make them look as weird as possible. A lot of my ideas have sprung from this. You scould also brainstorm bits and pieces of characters you like, such as: I love Kyou-kun's hair. You could take some elements you like, and mix them into a character. Just don't make it TOO similar, or you'll get accused of ripping off a character.
Good luck! Hope I could help!

And try to makes it fit.
